γ-Secretase in skin diseases

Xiao Xuemin, He Yanyan, Zhang Xiaofeng��Li Chengrang��Wang Baoxi��Institute of Dermatology, Chinese Academy of Medical Sciences and Peking Union Medical College��Nanjing 210042, China Corresponding author: Wang Baoxi, Email: wangbx@ncstdlc.org ��Abstract�� γ-Secretase, a transmembrane protease complex composed of four hydrophobic subunits, is capable of cleaving more than 30 type 1 transmembrane proteins such as β-amyloid precursor protein and Notch receptors. Recently, heterozygous mutations in the γ-secretase gene have been reported in multiple cases of familial and sporadic acne inversa. More and more data have highlighted γ-secretase to have a significant role in the maintenance of physiological function of normal skin and in the development of multiple skin diseases such as inflammatory dermatoses, skin tumors, infectious dermatoses, sexually transmitted diseases and connective tissue diseases. Although there has been no clinical practice, γ-secretase inhibitors and activators may open a new door in the prevention and treatment of skin diseases in the future.
