Roles of signal transducer and activator of transcription 3 and vascular endothelial growth factor in cutaneous melanoma: an update

Abstract:

YIN Fang*�� SONG Ning-jing, WANG Xiao-nan. *Shanghai Skin Disease Hospital, Anhui Medical University, Shanghai 200443, China Corresponding author: SONG Ning-jing, Email:songnj-10@163.com. ��Abstract�� Signal transducer and activator of transcription 3 ��STAT3��, known as an important member of the STAT family and the focus of many carcinogenic pathways, is involved in the regulation of gene expression, and its continuous activation is associated with the development of tumors. STAT plays an important role in cell proliferation, survival, apoptosis, differentiation and metastasis by modulating some regulatory factors, such as cell apoptosis-inhibiting factors��Bc1-xL, Bc1-2, survivin��, cell cycle-regulating factors ��cyclin-D1, cyclin-D2, c-myc�� and angiogenesis-inducing factors ��vascular endothelial growth factor ��VEGF��, cyclooxygenase 2 ��COX-2��, matrix metalloproteinase-9 ��MMP-9����. VEGF is now believed to be the most important angiogenic factor with a considerable effect on both vascular stroma formation and biological behavior of tumors, and can serve as a marker of tumor metabolism and metastasis. This paper elucidates the function and possible mechanisms of STAT3, VEGF and relevant regulatory genes in the initiation and progression of cutaneous melanoma, which may provide new ideas for the targeted therapy of cutaneous melanoma. ��Key words�� Melanoma; STAT3 transcription factor; Vascular endothelial growth factor A
