Safety Evaluation of Tumor Necrosis Factor Inhibitory Agents in the Treatment of Psoriasis

Abstract:

In recent years, a growing number of tumor necrosis factor inhibitory agents have been used in the treatment of psoriasis. They have shown good effects in the treatment of psoriasis; nevertheless,a variety of adverse reactions have been observed. Safety issues related to these agents include the risk of injection-site reactions or infusion-related reactions, infection, malignancy, autoimmune diseases, and of demyelinating disorders. Injection-site and infusion related reactions are most likely to be easily managed and rarely lead to discontinuation of therapy. The occurrence of infections can be decreased or prevented by screening, carefully monitoring and follow-up. More studies are needed to evaluate the risk of malignancies caused by these agents in the treatment of psoriasis. It is still not clear about the relationship between demyelinating diseases and these agents. Overall, these agents are well tolerated and have demonstrated favorable benefit-to-risk profile in the treatment of psoriasis. The adverse reactions, contraindications,and safety in pregnancy of these agents are reviewed.
