IL-12 and, more recently, IL-23 have been implicated in the pathogenesis of psoriatic lesions. New therapies, including two monoclonal antibodies against a subunit p40 shared by IL-12 and IL-23, namely ustekinumab and ABT-874, have been developed to treat psoriasis. Current data have shown that these agents exert an excellent efficacy in the treatment of psoriasis. However, long-term observations may be needed to assess their potential for infection and other adverse events.
